Like the title says in what scene across all four movies (including alternate versions) do you think he did his best work with the all around best support?

That's a tough one for STM- too many solo scenes... but...with 'best work' I assume you're talking performance-wise:

#1: The scene where Superman is about to reveal his identity to Lois, but backs off
#2: The scene where Lois dies/Superman screams to the heavens

SII:

#1: The diner scene
#2: The talk with Lois outside the Fortress of Solitude/on the balcony

SIII:

#1: The 'evil' Superman as he battles himself- he had to have had a blast playing that-

SIV:

#1: The speech at the United Nations. It could have come off corny, without true commitment to the material.

IMHO as an encapsulating moment:) :

After having been saved ,  Ricky exclaims "Superman!" ...and Reeve retorts with that classic smile: "that's me".

Sums it all up for me.

But there are loads more--lol
